骨形態(tài)發(fā)生蛋白(Bone Morphogenetic Protein,BMP)屬轉化生長因子超家族成員,這種蛋白質具有使未分化間充質定向分化為成骨細胞,并進而合成膠原、形成鈣化骨組織能力,因此命名為骨形態(tài)發(fā)生蛋白。BMP-4作為BMPs家族成員之一,在空間結構上與BMPs大體結構有一致性,只有在成熟區(qū)序列方面有其特異性。BMP是一種二聚體分子,由兩個單體以一個二硫鍵結合而成。它首先被合成為較大前體蛋白,前體含一個100-125個氨基酸組成羧基末端。通過蛋白質分解過程釋放后,兩個前體以二硫鍵結合發(fā)生二聚作用,有活性同二聚體或異二聚體被分泌出來。BMP-4氨基末端含有一段信號肽序列,且含有4個可能氨基末端連接糖基化位點,其中3個在同一位置。
人骨形態(tài)發(fā)生蛋白-4重組蛋白是在大腸桿菌中重組表達的,是一條單鏈、非糖基化的多肽鏈,包含有116個氨基酸,分子量為13009Da。該人骨形態(tài)發(fā)生蛋白-4重組蛋白最終由標準的色譜技術純化出。

Recombinant Human Bone Morphogenetic protein-4 【BMP 4】重組人骨形態(tài)發(fā)生蛋白-4 細胞因子產品使用須知: 
溶解:提供的人骨形態(tài)發(fā)生蛋白-4(BMP 4)產品為過濾滅菌的白色凍干粉末;該凍干粉末是從含1mg/ml人骨形態(tài)發(fā)生蛋白-4(BMP 4)的溶液(不含添加劑)中凍干產生的。我們建議加無菌的20mM AcOH(乙酸)以配制工作儲存液到不低于100ug/ml,后繼實驗過程中可根據實驗要求進一步稀釋。
保存:人骨形態(tài)發(fā)生蛋白-4(BMP 4)凍干粉末在室溫可保存3周,或置于-18℃凍存用于長期保存。重新溶解后可以在4℃冷藏保存至少2-7天以上;如果想延長保存時間,建議分裝(加載體蛋白如0.1% HSA或 BSA)后置于-18℃凍存。請避免反復凍融。

Introduction 

The protein encoded by this gene is a member of the bone morphogenetic protein family which is part of the transforming growth factor-beta superfamily. The superfamily includes large families of growth and differentiation factors. Bone morphogenetic proteins were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. This particular family member plays an important role in the onset of endochondral bone formation in humans, and a reduction in expression has been associated with a variety of bone diseases, including the heritable disorder Fibrodysplasia Ossificans Progressiva. Alternative splicing in the 5' untranslated region of this gene has been described and three variants are described, all encoding an identical protein.


Description 

Bone Morphogenetic Protein-4 Human Recombinant produced in E.Coli is a monomeric, non-glycosylated, Polypeptide chain containing 116 amino acids and having a molecular mass of 13kDa.
The BMP-4 is purified by proprietary chromatographic techniques.

Physical Appearance  (物理特征)

Sterile Filtered White lyophilized (freeze-dried) powder.


Formulation 

BMP-4 was lyophilized from a 0.2?m filtered concentrated (1mg/ml) solution in 20mM Na2CO3 buffer, pH 9.0.
Solubility  It is recommended to reconstitute the lyophilized Bone Morphogenetic Protein-4 in sterile 18M-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.


Stability  (穩(wěn)定性)

Lyophilized Bone Morphogenetic Protein-4 although stable at room temperature for 3 weeks, should be stored desiccated below -18°C. Upon reconstitution BMP4 should be stored at 4°C between 2-7 days and for future use below -18°C.
For long term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.

Greater than 95.0% as determined by:
(a) Analysis by RP-HPLC.
(b) Analysis by SDS-PAGE.
